Directed differentiaton of human embryonic stem cells into transplantable hepatocytes: Recent progress and Fugure directions

Abstract
Liver transplantation has been performed for the treatment of irreversible liver dysfunctions, including cirrhosis and fibrosis. However, this therapeutic option is limited due to the scarcity of donor livers. Hepatocyte transplantation has been suggested as an alternative application to whole organ transplantation, but availability of human liver cells is still a major obstacle to the cell replacement therapy. Human embryonic stem cells (hESCs) have been proposed as potential sources of hepatocytes based on their unlimited proliferation capacity and developmental potential to give rise to a variety of cell types, including hepatocytes. Here, this manuscript summarize recent progress in the directed differentiation of pluripotent stem cells into transplantable human hepatocytes and discuss about the current problems and future directions for clinical applications of the stem cell-based therapy for liver diseases.
